INTRODUCTION
PREMARIN 0.3MG contains Conjugated estrogens which belongs to the group of medicines called Hormone Replacement Therapy. It is used among postmenopausal women to relieve menopausal symptoms such as hot flushes, sleep problems, irritability, and depression.
It can also be used to prevent osteoporosis (bone loss and fragility) among postmenopausal women who are at risk of developing fractures easily. PREMARIN 0.3MG should be used only in women who do not have their period at least for the past 6 months.
Conjugated estrogen is a synthetic derivative of a female sex hormones called estrogen. It supplements our body with sufficient levels of estrogens which usually becomes deficient during menopause. As a result, it provides relief from postmenopausal symptoms such as hot flushes, sleep problems, irritability, depression, and bone loss.
PREMARIN 0.3MG is not a contraceptive (birth control pill), therefore it cannot prevent you from becoming pregnant. Women who are in pre-menopause (less than 6 months from last periods) or aged below 50 years must use a suitable contraceptive method to prevent pregnancy.
During management with PREMARIN 0.3MG, your doctor will instruct you to have periodic blood tests and regular physical examination (breast examination) to avoid the risk of unwanted side effects. Avoid drinking grapefruit juice while taking PREMARIN 0.3MG as it may interfere with the effects of PREMARIN 0.3MG.
Do not take PREMARIN 0.3MG if you have or previously had breast cancer, uterine cancer, unexplained vaginal bleeding, heart problems or blood clots. Before taking PREMARIN 0.3MG inform your doctor if you have any liver or kidney problems, diabetes, migraine headache, asthma, or epilepsy.
PREMARIN 0.3MG is not intended for use in pregnant and breastfeeding women. The most common side effects of taking PREMARIN 0.3MG are stomach pain, headache, vaginal infection, or vaginal discharge, weight gain, memory loss and changes in libido. Consult your doctor if any of these side effect worsens.
Also, inform your doctor if you experience any changes in your breast such as breast pain, tenderness, discharge from the nipple, or lump growth in the breasts, heavy menstrual bleeding, pain, swelling, redness of the legs, sudden chest pain or difficulty in breathing.

HOW PREMARIN 0.3MG WORKS
PREMARIN 0.3MG supplements our body with sufficient levels of female sex hormones (estrogen). This helps in manageing problems associated with hormonal deficiency states such as menopause. As a result, it provides relief from postmenopausal symptoms such as hot flushes, sleep problems, irritability, depression, and bone loss.
DIRECTIONS FOR USE
Take PREMARIN 0.3MG strictly as advised by your physician. Swallow PREMARIN 0.3MG with a glass of water. Do not crush or chew the medicine. Try to take PREMARIN 0.3MG at the same time each day.
Do not miss any dose of PREMARIN 0.3MG in between the therapy, as it may reduce its effectiveness. In case if you have missed a dose of PREMARIN 0.3MG take it as soon as you remember. Avoid taking a double dose of PREMARIN 0.3MG to compensate the missed dose it may lead to bleeding or spotting. Your doctor will decide the correct dose and duration of therapy for you depending upon your age, body weight and health condition.
SIDE EFFECTS OF PREMARIN 0.3MG
COMMON
- irregular menstrual bleeding
- vaginal infection (thrush)

- joint pain, leg cramps

RARE
Stop taking PREMARIN 0.3MG and contact your doctor immediately if you experience any of the following side effects:
- unexplained vaginal bleeding
- signs of abdominal uterine bleeding (such as changes in menstrual flow, breakthrough bleeding or spotting, pelvic pain, vaginal inflammation, and vaginal discharge)
- signs of blood clots such as pain, swelling and redness of the legs, sudden chest pain, and difficulty in breathing
- signs of liver problems (such as yellowing of the skin or the whites of the eyes (jaundice))
- signs of high blood pressure (such as headache, tiredness, dizziness)
- migraine-like headache happening for the first time
- signs of allergic reaction (such as skin rash, itching, shortness of breath, difficulty in breathing, and a swollen face)
- inflammation of the colon (part of the intestine) which may present as lower left sided abdominal pain and/or bloody diarrhea
- growth of benign meningioma (a tumor of the membranes around the brain or spinal cord)
- signs of breast cancer (breast pain or tenderness, swollen breasts, discharge from the nipples and changes in breast tissue)
HOW TO MANAGE SIDE EFFECTS
Nausea and Vomiting:
Take PREMARIN 0.3MG with or just after meals. Stick to simple meals. Avoid eating oil rich or spicy foods. Consult your doctor if your nausea worsens.
Headache:
Rest and relax. Keep yourself hydrated by drinking plenty of fluids such as water or electrolytes. Applying a pain-relieving balm on your forehead can be beneficial. Do not consume excessive alcohol, as it could aggravate your headache. Consult your doctor if your headache worsens.
Dizziness:
Try to rest and relax. Get enough sleep. Avoid driving or operating any tools or machines completely while you are feeling dizzy. Do not consume excessive alcohol, as it could aggravate your dizziness. Consult your doctor if your dizziness worsens.
WARNING & PRECAUTIONS
PREGNANCY
PREMARIN 0.3MG is not intended for use in pregnant women. However, if you are in pre-menopause (less than 6 months from last periods) or aged below 50 years, you must use a suitable contraceptive method to prevent pregnancy. Stop taking PREMARIN 0.3MG and consult your doctor immediately if you accidentally become pregnant while taking PREMARIN 0.3MG.
BREASTFEEDING
PREMARIN 0.3MG is not intended for use in breastfeeding women. Consult your doctor before taking.
DRIVING AND USING MACHINES
PREMARIN 0.3MG is unlikely to affect your ability to drive or to use machines. However, do not drive or operate any heavy tools or machines if your ability is affected by PREMARIN 0.3MG.
ALCOHOL
Avoid consumption of alcohol while taking PREMARIN 0.3MG, as it may increase the risk of side effects.
KIDNEY
PREMARIN 0.3MG should be used with caution in patients with kidney diseases. Therefore, consult your doctor before taking it.
LIVER
PREMARIN 0.3MG is not recommended for use in patients with severe liver diseases (Ex. benign liver tumor). Therefore, consult your doctor before taking it.
ALLERGY
Do not take PREMARIN 0.3MG if you are allergic to Conjugated estrogens.
LUNGS
PREMARIN 0.3MG is not recommended for use in patients having blood clots in their lungs (pulmonary embolism). It should be used with caution in patients having asthma. Therefore, consult your doctor before taking it.
HEART DISEASE
PREMARIN 0.3MG is not recommended for use in patients with heart problems (as heart attack). Therefore, consult your doctor before taking it.
OTHERS
PREMARIN 0.3MG is not recommended for use if you:
- have or previously had breast cancer
- have cancer in the uterus lining (endometrium), or if you are suspected of having it
- have any unexplained vaginal bleeding
- have unmanageed endometrial hyperplasia (excessive thickening of the layer of uterus)
- have or had blood clot in the veins (thrombosis, deep venous thrombosis, pulmonary embolism)
- have a blood clotting disorder (such as Protein C, Protein S, antithrombin deficiency)
- have porphyria (inherited disorder affecting RBC)
Before taking PREMARIN 0.3MG, inform your doctor if you:
- have fibroids (tumors growth in uterus)
- have endometriosis (a condition where inner lining of the uterus grows outside the uterus)
- have or had family history of breast cancer
- have diabetes
- have migraine (a neurological disease with symptoms of severe headache)
- have or ever had systemic lupus erythematosus (a serious disease in which immune system attacks its own body)
- have or ever had epilepsy
- have fluid retention
- have hypocalcemia (low blood calcium levels)
- have high blood fat levels
- have premature menopause
- have any blood tests to be done
- are overweight
- have or ever had hereditary and acquired angioedema
- have or ever had otosclerosis (disease affecting the eardrum and hearing)
- have any upcoming surgery
Use in pediatrics:
PREMARIN 0.3MG is generally not recommended for use in children and adolescents (aged below 18 years of age). Therefore, consult your doctor before taking it.
Use in geriatrics:
PREMARIN 0.3MG should be used with caution in elderly patients. Therefore, consult your doctor before taking it.
INTERACTIONS
A. Drug-Drug Interaction:
Before taking PREMARIN 0.3MG, inform your doctor, if you are taking any of the following:
- medicines used to manage epilepsy (Ex. phenobarbital, phenytoin, carbamazepine)
- medicines used to manage tuberculosis (Ex. rifampicin, rifabutin)
- medicine used to manage HIV infection (Ex. nevirapine, efavirenz, ritonavir and nelfinavir)
- medicines used to manage bacterial infections (Ex. erythromycin, clarithromycin)
- medicines used to manage fungal infections (Ex. ketoconazole, itraconazole)
- St. john’s wort (an herbal medication used to manage depression)
- metyrapone (a medicine used to adrenal insufficiency)
- cimetidine (a medicine used to manage stomach ulcers)
- dexamethasone (a corticosteroid used to manage inflammation)
B. Drug-Food Interaction:
Avoid drinking grapefruit juice while taking PREMARIN 0.3MG as it may alter the effects of this medicine.
Overdosage:
If you or anyone else accidentally took more PREMARIN 0.3MG, consult your doctor immediately or visit the nearby hospital. Symptoms of overdose might include nausea, breast tenderness, dizziness, stomach pain, drowsiness, weakness, and short periods of vaginal bleeding.
